Cytotoxicity assays measure loss of some cellular or intracellular structure and or functions, including lethal cytotoxicity. Cytotoxicity assays are widely used in fundamental research and drug discovery to screen libraries for toxic compounds.

Cytotoxicity assays are the cornerstone of any drug discovery project since they report on cell proliferation and cell metabolism state. These assays are applied extensively in any cancer research projects.For example, growth factor receptors (GFRs) are known for their involvement in cell growth and division, and cytotoxicity assays allow testing numerous drugs targeting GFRs via the measurement of the living (or dead) cells amount.

WE offer cytotoxicity cell-based assays measuring the amount of the living cells.

AlamarBlue

  • Assay reagent contains resazurin, an oxidation-reduction indicator

  • AlamarBlue is reduced by reactions connected to cell metabolism

  • Fluorescence intensity is proportional to the number of living cells respiring

  • Resazurin is non-toxic,so follow-up assays can be performed on same cells

MTT

  • MTT, a yellow tetrazole, is an oxidation-reduction indicator

  • MTT reduction to purple formazan is connected to NAD(P)H-dependent oxidoreductase enzymes activity

  • NAD(P)H flux depends on cell growth and division

  • Resulting absorbance of the coloured solution is measured at 570 nm wavelength

CellTiter-Glo

  • ATP acts in synergy with events important for cell’s growth, division and metabolism

  • CellTiter-Glo assay is homogenous, simply added, mixed and measured

  • Luciferase reaction used in this assay requires ATP to produce light

  • The amount of light is proportional to the number of living cells

CytoTox

  • LDH is an enzyme released upon cell lysis

  • LDH plays a role in the conversion of a tetrazolium salt (INT) into a red formazan

  • The amount of coloring is proportional to the amount of lysed cells

  • CytoTox allows measuring cell death triggered by chemicals, proteins, bacteria, viruses, etc.
